Regeneration of Degenerated Intervertebral Disc with Autologous Bone Marrow Mononuclear Cells in Rabbit Models

Abstract

Objective To investigate the regenerative effect of autologous bone marrow mononuclear cells on degenerated intervertebral disc in rabbit models.

Methods A total of 12 New Zealand white rabbits were selected. The rabbits were randomly divided into two groups: the control group (n=6) and the treatment group (n=6). Bone marrow was extracted from the posterior superior iliac spine of the New Zealand white rabbit, and autologous bone marrow mononuclear cells were extracted by density gradient centrifugation. L4/5, L5/6 and L6/7 intervertebral disc puncture models were made with 16 G needle after anesthesia in the two groups of rabbits. Autologous bone marrow mononuclear cells were injected into the punctured disc of the treatment group and no treatment was done in the control group. Then the rabbits in the two groups were raised under the same conditions. Imaging (X-ray and MR) and histological observation of the disc were performed at weeks 4, 8, and 24, respectively.

Results In terms of maintaining the height of intervertebral space, the treatment group was superior to the control group as the number of weeks increased. In terms of signal strength of MR T2W1 and histological morphology, the treatment group was significantly better than that of the control group. Histological observation showed that compared with the treatment group, the number of nucleus pulposus cells in the control group was reduced, and the extracellular matrix was contracted and uneven.

Conclusion Bone marrow monoculear cells transplantation can regenerate the degenerated intervertebral disc in rabbit models.
