The Effect of IFN-γ Increasing MHC-Ⅱ Expressing to the Immunogenicity of Keratinocytes

Abstract: Objective To observe changes of immunogenicity of keratinocytes(KC) under the influence of interferon-γ(IFN-γ).Methods KC were cultured and passaged in vitro.Different dose of IFN-γ stimulated KC to express MHC-Ⅱ molecules.KC with highty expressed MHC-Ⅱ molecules were mixed with allogenic lymphocytes to observe their proliferation.Simultaneously,MHC-Ⅱ+ KC were implanted subcutaneously into allogenic mice to observe the sign of immunological rejection.Results Under the influence of 6 000 U/mL IFN-γ,the percentage of MHC-Ⅱ+ KC exceeded 90%.MHC-Ⅱ molecules highly expressed KC had no effects to stimulate proliferation of allogenic lymphocytes significantly.There were no obvious lymphocytes infiltration when MHC-Ⅱ molecules highly expressed KC were implanted subcutaneously into allogenic mice.Conclusion Passaged KC could express MHC-Ⅱ molecules highty in the influence of IFN-γ,KC expressed MHC-Ⅱ molecules has no obvious immunogenicity.
