Abstract

Objective To explore the feasibility of constructing tubular cartilage with a diameter similar to goat tracheal in vitro by large mammalian auricular cartilage, based on improved cell-sheet technology. Methods The chondrocyte cell-sheet was cultured for 1-2 weeks in vitro, and was shaped as tubular cartilage tissue.Samples were collected at 6 weeks. The general appearance and elasticity were observed, and the formation of cartilage was examined. Results After cultured for 6 weeks in vitro, the cell-sheet had intergraded to a white translucent tubular tissue. The tubular tissue had certain biomechanical strength similar to mature cartilage tissues, and the histology analysis displayed continuous cartilage phenotype. Conclusion The improved cartilage cell-sheet technology has a series of advantage including limited defect in donor site, short period in vitro culture, less inflammatory response in vivo and so on.This technology would provide a promising approach for the regeneration of cartilage in vivo.
