A Study of Constructing Tubular Tissue Engineered Cartilage by Using Chondrocytes Cell-Sheet Approach

Abstract: Objective To explore the feasibility of constructing tubular cartilage with a diameter similar to goat tracheal in vitro by large mammalian auricular cartilage,based on improved cell-sheet technology.Methods The chondrocyte cell-sheet was cultured for 1-2 weeks in vitro,and was shaped as tubular cartilage tissue.Samples were collected at 6 weeks.The general appearance and elasticity were observed,and the formation of cartilage was examined.Results After cultured for 6 weeks in vitro,the cell-sheet had intergraded to a white translucent tubular tissue.The tubular tissue had certain biomechanical strength similar to mature cartilage tissues,and the histology analysis displayed continuous cartilage phenotype.Conclusion The improved cartilage cell-sheet technology has a series of advantage including limited defect in donor site,short period in vitro culture,less inflammatory response in vivo and so on.This technology would provide a promising approach for the regeneration of cartilage in vivo.
