大鼠BMSC 与ADSC 体外Transwell 共培养诱导成骨的实验研究

Experimental Study on the Osteogenic Induction by Transwell Co-Culture of Rat BMSCs and ADSCs in Vitro

摘要: 目的研究大鼠骨髓基质细胞(Bone marrow stromal cells,BMSCs)与脂肪来源基质细胞(Adipose-derived stromal cells,ADSCs)体外Transwell共培养后对于ADSCs成骨分化的影响。方法取来源于同一只6周龄SD雄性大鼠的ADSCs及BMSCs,培养传代至第2代后按1∶1比例共培养(共培养组),下室植入ADSCs,上室植入BMSCs;同时设置单纯ADSCs组及BMSCs组为对照组。21 d时成骨诱导分化,然后行茜素红染色,比较共培养后BMSCs对于ADSCs成骨分化的影响。结果共培养组茜素红半定量测定结果与ADSCs组相比有显著性差异(P0.05)。结论 ADSCs和BMSCs体外Transwell共培养后,其成骨矿化能力增强,BMSCs可通过旁分泌方式促进ADSCs成骨分化。

Abstract: Objective To study the effect of Transwell co-culture of rat BMSCs and ADSCs on the osteogenic differentiation of ADSCs in vitro. Methods ADSCs and BMSCs were obtained from the same 6-week-old SD male rats. The second passage of ADSCs and BMSCs were harvested and placed into Transwell co-culture system at a ratio of 1 ∶1(co-culture group), ADSCs were placed in the bottom and BMSCs in the upper. Meanwhile ADSCs group and BMSCs group were served as control group. After 21 days of osteogenic induction and differentiation, the qualitative and semi-quantitative staining analysis of alizarin red were used to investigate the effect of BMSCs on ADSCs osteogenic differentiation. Results The results of semi-quantitative determination of alizarin red in the co-culture group were statistically different from that in the ADSCs group(P0.05). Conclusion Osteogenic mineralization ability was enhanced after in vitro Transwell co-culture of BMSCs and ADSCs, and BMSCs could promote osteogenic differentiation of ADSCs by paracrine.
