Preliminary Study of SVF-gel Promoting Wound Healing in Diabetic Rats

Abstract: Objective To investigate the effect and mechanism of SVF-gel promoting wounds healing in diabetic rats.Methods Thirty-six male SD rats were prepared by intraperitoneal injection of Streptozotoein(55 mg/Kg)to establish the diabetic model.The wound with a diameter of 0.6 cm was made on both sides of diabetic rats after one week.Then those rats were divided into 3 groups(n=12)randomly,and treated with SVF-gel(Group A),SVF(Group B),and normal saline(Group C)respectively.The wound sites were observed and the percentage of wound closure was calculated.HE staining and immunohistochemical staining were used to observe the morphological changes of granulation tissue in the wound and the angiogenesis.Results The wound healing rates at all time points of group A and group B were higher than that of group C(P<0.05),and the wound healing rates at all time points of group A were higher than that of group B(P<0.05).Histological changes:Compared with group C,the infiltration of inflammatory cells was decreased in group A and group B,the degree of reepithelialization was obvious,the angiogenesis was significantly increased,the granulation tissue structure was dense,and these histological changes were the most obvious in group A.Conclusion For the wounds of diabetic mice,SVF-gel may increase the formation of blood vessels and the granulation tissue,reduce the inflammatory reaction to promote the wound healing.
