Abstract: Objective To explore the efficacy of repairing skin defect of mouse by using embryonic skin cell. Methods Animal model of back skin damage in mice was built. The embryonic skin cells were transplanted into the back skin damage of mice as treatment group (Nor-T group, n=6) and the mice in control group (Nor group, n=6) were received no treatment. The wound healing situation were observed 1, 2 and 3 weeks after operation. The expression of TGF-βwas detected 1 and 2 weeks after operation. Results The regenerative skin growth was observed on the wound area in Nor-T group, while only crusta was observed on the wound area in Nor group 1 week after operation. The regenerative skin merged with surrounding skin in Nor-T group, while the wound healed with scar in in Nor group 2 weeks after operation. The hair was observed in Nor-T group, while no hair was observed in Nor group 3 weeks after operation. In Nor-T group, TGF-β1 was expressed 1 week after operation and TGF- β3 was expressed 2 week after operation. Conclusion The embryonic skin cells possess the ability of regenerative skin formation, and TGF-βhas different expression at different wound healing phase.
