摘要: 目的 探究BK通道抑制剂对人脐带华通胶来源的间充质干细胞(WJ-MSC)的细胞活性、细胞凋亡、细胞因子分泌的影响。方法 以组织贴壁法分离培养人WJ-MSC,Western-blot检测BK通道的表达;用不同浓度BK通道抑制剂处理后,CCK-8法检测细胞活性,Annexin-V-FITC/PI检测细胞凋亡,Elisa检测细胞因子(IL6、IL10)的表达。结果 人WJ-MSC上存在BK通道的表达,BK通道抑制剂能够明显降低细胞活力(P<0.05);促进细胞凋亡(P<0.05);促进细胞因子(IL6)的分泌(P<0.05),但并不增加IL10的分泌。结论 BK通道抑制剂能够抑制人WJ-MSC的活性,促进细胞凋亡,促进IL6的分泌。

Abstract: Objective To explore the effects of BK channels inhibitors on cell activity,apoptosis and cytokine secretion of mesenchymal stem cells derived from human umbilical cord Wharton's jelly(WJ-MSC).Methods The WJ-MSC were isolated and cultured by tissue adherent method.The expression of BK channels was detected by Western-blot.After treated with different concentrations of BK channels inhibitors,the activity of cells was detected by CCK-8,the apoptosis of cells was detected by Annexin-V-FITC/PI,and the expression of cytokine(IL6,IL10)was detected by Elisa.Results BK channels expression was found in WJ-MSC.BK channels inhibitors reduced cell activity,promoted cell apoptosis and secretion of cytokine(IL6),all of which have statistical significance(P<0.05).But BK channels inhibitors did not increase the secretion of IL-10.Conclusion BK channels inhibitors can inhibit the activity of WJ-MSC,promote cell apoptosis and promote the secretion of IL6.
