Isolation of Specific Sequence of TGF-β Receptor Ⅱ Using a Phage 12-mer Display Peptide Library

Abstract: Objective To isolate the specific sequence of TGF-β receptor Ⅱ(TβRⅡ) from a phage 12-mer display peptide library and to evaluate their effects on keloid fibroblasts.Methods A phage 12-mer display peptide library was screened for four rounds using monoclonal anti-human TGF-β1 as the target.ELISA was performed to select monoclonal phages with good association for DNA sequencing.MTT assay was used to detect the effects of monoclonal phages on keloid fibroblasts.Results Five specific sequences were obtained,which were similar to TβRⅡ.The results of MTT assay showed that three of the five phage model peptides could inhibit proliferation of keloid fibroblasts.Conclusion Phage model peptides with specific sequences of TβRⅡ can inhibit proliferation of keloid fibroblasts.
