    Preliminary Study on Inducing Hair Follicle Stem Cells into Vascular Smooth Muscle Cells in Vitro

    Objective To investigate the feasibility of inducing human hair FSCs(follicle stem cells) into VSMCs(vascular smooth muscle cells) in vitro.Methods The human hair FSCs were isolated by dispase and cultured in DMEM containing 10 ng/mL PDGF-BB as experimental group,and the human hair FSCs cultured in DMEM without PDGF-BB were set up as control group.The morphological change of each passage was observed,the expression of α-Smooth Muscle actin(α-SM actin) and calponin in each group were tested after inducing FSCs at passage 4.Results The VSMC-like change was obviously observed in the induced FSCs,while the same phenomenon was not found in control group.α-SM actin and calponin were expressed by immunocytochemical staining in the induced FSCs of fourth passage,while little expression was observed in the control group.FCM showed nearly 50% of expression rate of α-SM actin and calponin in the experimental group,much higher than 8% in control group.RT-PCR further confirmed the expression of calponin in experimental group,and no obvious expression of calponin was tested in control group.Conclusion FSCs can be induced into VSMCs in vitro by DMEM containing 10 ng/mL PDGF-BB.

    Effects of Various Concentrations of Glucose and Glutamine on the Cultivation of Human Umbilical Cord-derived Mesenchymal Stem Cells in Vitro

    Objective To explore the effects of various concentrations of glucose and glutamine on the cultivation of human umbilical cord-derived mesenchymal stem cells(hUC-MSCs) in vitro.Methods The hUC-MSCs were separated and cultured.The morphology and surface antigens were investigated by optical microscopy and flow cytometry.Then hUC-MSCs were cultured with various concentrations of glucose and glutamine in vitro.Cells on microcarriers were counted by crystal violet staining.The nutrients and their by-products in the medium supernatant were measured.Results The markers of CD29,CD44,CD105,and CD90 were strongly expressed in hUC-MSCs,but the markers of CD14 and CD45 were not.With the increase of glucose concentration,the expansion of cells increased,glucose consumption,lactate formation and glutamine consumption had no change,and the ammonium formation improved.The increase of glutamine concentration had a limited effects to improving cell proliferation,but it shorted the culture period as improving growth rate.The increase of glutamine concentration had no effect on the consumption of glucose and glutamine,but improved the formation of lactate and ammonium.Conclusion Glucose and glutamine both enhanced cell proliferation.Lack of glucose results in the larger consumption of amino acids to provide nitrogen,and large supply of glutamine leads to the larger formation of lactate from glutamine.

    The Study of Combination of Human Adipose-Derived Stem Cells with Collagen Scaffold

    Objective To observe the growth of human adipose derived stem cells co-culture with collagen biomaterials in vitro and in vivo.Methods hADSCs were isolated from human subcutaneous adipose tissue after collagenase digesting,filtrating and centrifuging.Then the 3 passage of hADSCs were seeded onto the collagen scaffolds and the growth of hADSCs co-culture with collagen biomaterials were observed by using Haematoxylin-Eosin staining,meanwhile the cells in the scaffolds cultured in vivo were detected by immunohistochemistry.Results Primary cultured hADSCs were spindle-shaped or fibroblasts-liked cells,and it could form the clone groups.Immunofluorescence identification of Vimentin was positive,and Flow cytometry results showed that 3 passage of ADSCs were positive for CD29,CD44,CD105,but negative for CD34,CD45.The histological study found that after cultured both in vitro and in vivo,hADSCs could grow in the space of the collagen scaffolds,moreover there were more cells in the scaffolds cultured in vivo than in vitro.When cultured in vitro,a few cells adhered at the edge of the collage 1 week later and more cells grew into the inside of the scaffolds after 2 weeks.When cultured in vivo,several cells were found at the edge of the scaffolds and parts of cells had grown into the inside even the whole layer of the scaffolds after 2 weeks,at the 4th week,a great deal of cells grew into the whole layer of the scaffolds.Simultaneously,immunohistochemistry results suggested the cells in the scaffolds’ inside cultured in vivo were from hADSCs.Conclusion Collagen scaffolds have a good biocompatibility with hADSCs,which can be used as a vehicle for hADSCs to repair tissue defect.

    The Study on Biological Change of BMSCs During the Osteogenesis of Tissue-Engineered Bone in Vivo: the Feasibility of CM-DiI as a Long-Term Tracer of BMSCs

    Objective To investigate the feasibility of CM-DiI on long-term tracing the biological change of bone mesenchymal stem cells(BMSCs) during osteogenesis of tissue engineered bone(TEB) in vivo,and to offer an alternative labeling method for tracing seed cells in vivo in future.Methods After BMSCs were separated from bone marrow in Beagle dogs and osteo-induced to the second generation(P2),the cells were labeled with the fluorescent dye CM-DiI.The morphologic change of BMSCs after labeling was observed by fluorescence convert microscope.The proliferative ability was tested by MTT colorimetric between the labeling and un-labeling groups.The expression of Collagen I,BMP-2,BGLAP,and SPARC were detected through RT-PCR between the two groups.Lastly,CM-DiI-labeling BMSCs and beta-TCP were co-cultured into TEB which was implanted subcutaneously into Beagle dogs.Eight weeks later,the implant was harvested and the biological change of BMSCs was observed by fluorescence convert microscope.At the same time,the ectopic osteogenesis of the TEB was tested by HE staining.Results Shortly after being labeled with CM-DiI,ringlike red fluorescence was present in the cytoplasm of BMSCs.48 hours later,the fluorescence was enhanced and was not weaken during 72 hours.The morphology of labeled BMSCs was consistent with unlabeled ones.There was no significant difference in the proliferative ability between the labeled and unlabeled groups.The expression of Col-Ⅰ,BMP-2,BGLAP,SPARC revealed that the CM-DiI labeled BMSCs were induced into osteoblasts successfully either.After seeding into β-TCP,the proliferation of labeled BMSCs was normal and abundant extracellular matrix(ECM) was observed in the pores of the ceramic by scanning electron microscope(SEM).After harvesting the composite of labeled cells and ceramic 8 weeks later,the cells with red fluorescence were observed by fluorescent microscope.HE staining revealed deposited osteoid in the pores of ceramic.Conclusion CM-DiI has no obvious impact on the growth,proliferation and osteogenic differentiation of BMSCs.The BMSCs in the composite of TEB could be traced as long as 8 weeks.It suggests a feasible method in monitoring of cells in vivo.

    The Effect of PRP Promoting the Expression of Collagen and Hyaluronic Acid of Light Aging Human Skin Fibroblasts in Vitro

    Objective To explore the effects of platelet-rich plasma(PRP) on collagen and hyaluronic acid(HA) expression of light aging human skin fibroblasts(HSFs) in vitro.Methods Cultured human skin fibroblasts were radiated with ultraviolet ray and the proliferation and doubling time of cell were estimated.Different concentrations(10%,30% and 50%) of PRP were added into cultured light aging human skin fibroblasts,and HSFs radiated with ultraviolet ray were taken as negative control and HSFs were taken as positive control.ELISA was used to measure collagen expression and hyaluronic acid expression.Results The cell proliferation was prohibited after ultraviolet irradiation and cell doubling time was shortened with the increasing of PRP concentration after PRP added.ELISA assay showed that the collagen type 1(COL1),type 3(COL3) and HA expression were decreased after ultraviolet irradiation and promoted after PRP added,significant difference was shown between expermental groups and negative control group(P<0.01).Conclusion PRP can promote the synthesis of collagen type 1(COL1),type 3(COL3) and HA,and may be used as an effective factor against skin photoaging.

    Clinical Application of Vacuum Sealing Drainage Combining with External Fixation Stents in the Treatment of Lower Limb Destroyed Damage

    Objective To explore the effect of the vacuum sealing drainage(VSD) combining with external fixation stents in the treatment of lower limb destroyed damage.Methods From October 2007 to November 2011,the VSD combining with external fixation stents was used in 22 patients with lower limb destroyed damage.All the patients were performed emergency debridement,open reduction,external fixation,and the wounds were covered by VSD materials with positive washing function.The repair of the wounds of second stage was performed by skin grafts or vascular pedicle flap when the wounds were clean and granulation growth was observed.Results Twenty-two patients were followed up for 12 to 24 months.All wounds were closed after VSD treatment for 7 to 21 days(average 13 days).Infection was occurred in 3 cases(13.6%),and was healed by daily dressing and antibiotic treatment.The fracture healing time was 4 to 12 months(mean 6.5 months).Conclusion The VSD combining with external fixation stents could stabilize the fracture effectively,early close the wounds,promote the fracture healing,reduce complications,and is a good choice for the treatment of lower limb destroyed damage.

    Effectiveness of 0.5% Solution of Topical Timolol Maleate for Infantile Hemangiomas

    Objective To investigate the clinical efficacy and safety of 0.5% solution of topical timolol maleate for infantile hemangiomas.Methods A total of 25 infants with hemangiomas were treated by 0.5% solution of timolol maleate topically every eight hours in a thin layer(30 μL/cm2) as treatment group,while 17 cases of infants with hemangiomas were treated with normal saline topically every eight hours in a thin layer(30 μL/cm2) as control group.The parents are responsible for topical application of drug every day.The treatment lasted for six months and the patients were visited once a month.The changes of tumor size,texture and color were monitored and recorded at a regular interval.Symptomatic treatment was given timely for any acute signs and symptoms.Results In treatment group,the overall response of excellent was evaluated in 1 patient,good in 4 patients,moderate in 11 patients,poor in 9 patients,and the total effective rate was 64.00%.In control group,the overall response of excellent was evaluated in 0 patient,good in 0 patient,moderate in 3 patients,poor in 14 patients,and the effective rate was 17.65%.There was significant difference between the two groups(P<0.05).No obvious adverse reactions were observed.Conclusion Topical timolol maleate shows good therapeutic effect for superficial hemangiomas with good drug tolerability and minor adverse reaction.

    Objective To explore the effect of functional reconstruction in the reconstruction of transverse facial cleft.Methods From 2003 to 2011,11 cases with transverse facial cleft were repaired by functional reconstruction.The skin of cheek was crossed by Z flap to prevent the scar from shrinking straightly and the thin tissues from depressed deformity.Functional muscle trimming was done to repair the ring structure of musculus orbicularis oris.Results All the patients were followed for 3 to 12 months and there were no angular chilitis and formation of ulceration.The natural appearance of the lips,coordinate movements and plump cheeks were achieved.Conclusion Functional reconstruction of transverse facial cleft could improve the coordinate lip movements greatly and achieve natural facial expression.

    Management of Soft-tissue Defect after the Surgery for Calcaneal Fractures with Lateral Approach

    Objective To investigate the efficacy of retrograde sural neurovascular flap in the repair of soft-tissue defect after the surgery for calcaneal fractures with lateral approach.Methods From September 2009 to December 2011,24 patients with soft-tissue defect after the surgery for calcaneal fractures with lateral approach were treated with retrograde sural neurovascular flap.The tissue defects ranged from 3.0 cm × 1.5 cm to 5.0 cm × 2.5 cm and were repaired with retrograde sural neurovascular flaps ranged from 4.0 cm × 3.0 cm to 6.0 cm × 4.0 cm in size.Sural nerve and local cutaneous nerve were anastomosed was done to reconstruct cutaneous sensation.Results All flaps survived completely without vascular problems.The donor site and wound were healed by first intention.All the patients were followed up for 6 to 15 months.The flap appearance was satisfactory.No pigmentation and contraction were observed.The two-point discrimination of the flap was 7-12 mm.Conclusion Using retrograde sural neurovascular flap is an ideal method in repairing the soft-tissue defect after the surgery for calcaneal fractures with lateral approach.

    Conservative Treatment of Orbital Fracture in Early Stage

    Objective To investigate the indications of conservative treatment of orbital fracture in early stage.Methods From October 2005 to December 2010,720 cases with orbital fracture were viewed.The indications of conservative treatment included: ①Negative in traction test;② No enophthalmos;③ The fracture area was less than 1.0 cm2.Other patients will be treated by surgical treatment.Results All cases were followed up for 1-12 months,there was no further ocular mal-position in 330 cases with conservative treatment.The ocular position,facial symmetry and visual function all return to normal in 390 cases with surgical treatment.Conclusion The indications of conservative treatment of orbital fracture in early stage suggested in this study may be useful and can avoid unnecessary operation.

    Objective To explore the effects of silica gel prosthesis combined with acellular allosome dermal pad in low nasal tip rhinoplasty.Methods While silica gel prosthesis rhinoplasty was performed on 82 patients with low nasal tip,acellularallosome dermal pad was sutured on the top of silica gel prosthesis,then placed into the cavity gap of the nasal prosthesis.The nasal tip was lifted attributed to the thickness of the silica gel prosthesis and the effect of dermal pad so as to achieve the purpose of raising the nasal tip.Results All the cases were followed up for 6 month to 2 years,the height of nasal tip had been greatly improved,low nasal tip had been corrected,and the results were satisfactory.Conclusion Silica gel prosthesis combined with acellular allosome dermal pad was effective in low nasal tip rhinoplasty,and is worthy of clinical promotion.

    LPHP and LC-DCP for the Treatment of Proximal Humeral Fractures in Patients with Different Age

    Objective To compare the clinical effects of locking proximal humeral plate(LPHP) and normal plate for the treatment of C2 type proximal humeral fractures in patients with different age.Methods Since January 2004 to June 2009,152 patients with C2 type proximal humeral fractures were divided into three age groups: youth(<30),middle-aged(30~60) and old(>60).Each age group was randomly divided into two groups: 77 cases in group A were treated by LPHP and 75 cases in group B were treated by normal plate.Fracture healing rate and Neer score were assessed to compare the clinical effects in different age groups of each group.Results All the patients were followed up for 18 to 24 months.There was no statistical difference in fracture healing rate between group A and group B in youth and middle-aged age groups(P>0.05),while in old age group,the fracture healing rate of group A(97.1%) was significantly higher than group B(81.8%)(P<0.05).According to Neer criteria,there was no statistical difference in Neer score between two groups in youth age group(P>0.05),but group A had clearly higher scores than group B in other two age groups(P<0.05).Conclusion Compared to normal plate,LPHP is much safer and more effective in the treatment of middle and old aged patients with C2 type proximal humeral fractures.

    Mesenchymal stem cell(MSC) has broad prospects in the field of regenerative medicine and tissue engineering due to its highly self-renewal and multi-lineage differentiation ability.Although there are many researches about MSC osteogenic differentiation,its osteogenic efficiency is still very low both in vitro and in vivo.Therefore,how to improve the MSC osteogenic performance has become the focus of researchers.MSC osteogenic performance is influenced by many factors.In this review,the influence factors are discussed from four aspects: cell sources,approaches of isolation and purification,induction strategies and vascularization.

    The nitric oxide-cyclic GMP(NO-cGMP) signaling system mediates important physiological functions associated with various integrative body systems.Massive researches show that NO –cGMP signaling plays an important role in inducing the stem cells into the myocardial cell,neuron,etc.Besides,NO – cGMP signaling can also promote the axonal elongation.This review focuses on the role of NO-cGMP signaling in the differentiation of stem cells such as embryonic stem cells(ESCs),mesenchymal stem cells(MSCs) and neural stem cells(NSCs).

    Mechanical stress-induced apoptosis,as programmed cell death,is often correlated with many diseases.Research of mechanical stress-induced apoptosis could provide the prevention and therapy of relative diseases.In this review,the basic features of apoptosis,relative research of stress-induced apoptosis,the establishment of the strain model and the methods of cell apoptosis detection are all reviewed.The research direction of stress-induced apoptosis is also proposed.
