    Purification of Vascular Cells with MACS and FACS for Constructing Vessel from Differentiated Mouse Embryonic Stem Cells

    Objective To compare the efficiency between using FACS and MACS for purification of smooth muscle cells and endothelial cells induced from differentiated mouse embryonic stem cells.Methods FLK-1+ cells were sorted from EBs by FACS and MACS,and were subsequently induced to vascular endothelial cells and smooth muscle cells with PDGF and VEGF respectively.Cells were then characterized by immunefluoresence staining and FACS.Results Large amount of vascular smooth muscle cells could be obtained from FLK-1 positive cells sorted by MACS.Over 90% of these cells were positive for smooth muscle markers,including SM α-actin,SM-SMC and Calponin.However,low output of smooth cells was observed by FACS sorting.Nearly 60% of cells expressed endothelial cell markers CD31 and vWF after VEGF induction of sorted cells by FACS,but low purity of endothelial cells was achieved from MACS sorting.Conclusion Large quantity of vascular smooth muscle cells with high purity can be obtained by MACS selection of FLK-1+ cells from EBs while high quality of vascular endothelial cells could be achieved by FACS sorting.

    The Effect of Tenomodulin Over-expression on the Expression of Tenocyte Related Molecules of NIH3T3 and C3H10T1/2 Cell Lines

    Objective To study the effect of tenomodulin(TNMD) over-expression on the expression of tenocyte related molecules of NIH3T3 and C3H10T1/2 cell lines.Methods The NIH3T3 and C3H10T1/2 cells were cultured in vitro and then transfected with pCAGGS vector and pCAGGS-TNMD expression vector respectively.After confirmation of successful transfection by RT-PCR,the morphology of transfected cells were observed with inverted phase contrast microscope,and the expression of some tenocyte related genes in transfected cells were analyzed with Quantitative PCR.Results There were no obvious changes in the morphology of NIH3T3 and C3H10T1/2 cells after transfection.The gene expression of collagenⅥ and biglycan in NIH3T3 cells decreased dramatically,while that of collagenⅠdid not shown obvious change.However,in C3H10T1/2 cells,the expression of collagenⅠand biglycan increased dramatically respectively,collagenⅥ did not shown obvious difference.Conclusion Tenomodulin(TNMD) over-expression of NIH3T3 and C3H10T1/2 cell lines did not affect the cell morphology,but could affect the expression of some tenocyte related genes.

    The Effects of Mesenchymal Stem Cells Transplantation on Survival of Prefabricated Flap in a Rabbit Model

    Objective To evaluate the effects of bone marrow-derived mesenchymal stem cells(BMSC) transplantation on survival of prefabricated flap with increased neovascularization in a rabbit model.Methods BMSCs of New Zealand rabbits were isolated and cultured in vitro.The third passage cells were labeled before transplantation.Two prefabricated flaps of each rabbit were randomly allocated to experiment group or control group.In all animals,the subcutaneous tissue around the tunnel was injected with BMSCs in experiment group,or saline in equal amounts in control group.One week later,one flap from each group was selected to observe the viability of BMSCs.Two weeks later,abdominal island flap based solely on the implanted vessel was elevated.Three flaps were selected from each group to observe the expression of VEGF by Western Blot.Four flaps were selected from each group to observe the signal of vWF and BrdU by immuno-fluorescence histochemical staining.Results BMSCs were found viable in the flap from the experiment group.There was a significant increase in mean survival rate of prefabricated flaps from the experiment group compared to the control group: BMSCs(93.1±2.6)% vs.saline(51.5±7.5)%(P<0.01).Immuno-fluorescence histochemical staining found the signal of vWF in the cytoplasm of BrdU positive cells.Western Blot semi-quantitative analysis showed that the The VEGF protein level was found significantly higher in the tissues from the experiment group than that from the control group.Conclusion The findings from this study demonstrated that BMSCs transplantation could promote neovascularization and improve prefabricated flap survival.

    Objective To investigate the effects of adipose derived stem cells(ADSC) on promoting lymphatic regeneration of lymphedematous mice tail.Methods ADSCs were isolated from the fat pad of the GFP transgenic mice and cultured to the second passage.Thirty days later,the ADSCs of the second passage were transplanted to the obstructed regions of the lymphedematous mice tail and the transplantation was repeated every seven days.The circumstance of tail were measured every third days and the lymph flow pattern was detected through micro-lymphangiography.Results ADSCs could participate in the formation of the crude pre-lymphatic fluid channels but no significant difference was observed on the reduction of the mice tail lymphedema.Conclusion The ADSCs could participate in the formation of pre-lymphatic fluid channels on the obstructive lymphatic regions,but there was no obvious effects on improving lymph flow.

    Objective To evaluate the effects of bone mesenchymal stem cells combining alginate on treatment of the degenerative intervertebral disc.Methods Fifteen New Zealand white rabbits were punctured in the vertebral gap to set up intervertebral disc degeneration model and were randomly divided into three groups,control group,blank group and experimental group.The rabbits were injected bone mesenchymal stem cell combining alginate gel stents in experimental group,alginate gel in control group,saline in blank group.The effect of repair was observed by MRI,immunohistochemical and biochemical analysis.Results Disc height,T2 value and the content of proteoglycan and Type Ⅱ collagen were higher in experimental group than in control and blank group.There was significant difference(P<0.05).Conclusion BMSC combining alginate could be used to repair the degenerative disc.

    Objective To evaluate the clinical effects of autologous bone marrow stem cells transplantation in treating diabetic arterial diseases of the lower extremities.Methods Thirty patients with diabetic artery diseases involved in both lower limbs were enrolled in this study.The patients were received autologenous bone marrow stem cell transplantation by intramuscular injections at multiple sites of affected limb.During 15 months of follow-up,symptoms including skin colour,sensation,local ulcers,skin temperature and claudication were observed and compared with the control group.The ankle-bronchial indexes and MRI angiographic results were recorded.Results The symptoms were greatly relieved and the walking distance without pain was dramatically prolonged.The ankle-bronchial indexes were increased from 0.35±0.20 to 0.56±0.10(P<0.05) after cellular treatment.Conclusion Autologous bone marrow stem cells transplantation is safe,effective and feasible for the treatment of peripheral arterial vascular disease in diabetic.

    Reconstruction of Postoperative Defects of Maxillary Sinus Using Free Anterolateral Flaps

    Objective To explore the clinical application of free anterolateral flaps in reconstruction of postoperative defects of maxillary sinus.Methods The postoperative defects of maxillary sinus were reconstructed using free anterolateral flaps in 17 cases.Results Among the 17 cases,2 cases occurred vascula crises,1 case occurred flap necrosis,5 cases had early local complications.After 3 months to 4 years follow-up,the appearance and functions on the head and heck area turned to be normal in 10 cases.No severe complication occurred in both donor and recipient site.2 patients survived with tumor and 7 patients died of local recurrence,cervical lymphatic metastasis and distant metastasis.Conclusion Anterolateral flap is easy to shape and has less complications of donor site.It is useful in the reconstruction of postoperative defects of head and neck tumor.

    The Prospective Study of Open Reduction and Internal Fixation of Proximal Humeral Fractures Using Locking Proximal Humerus Plate

    Objective To evaluate the functional outcome and the complication rate after open reduction and internal fixation of proximal humeral fractures using locking proximal humeral plate.Methods One hundred and six patients(mean age,62.9±15.7 years) with acute proximal humeral fracture were managed with open reduction and internal fixation using locking proximal humeral plate.With the three-month,six-month,and one-year follow-up,all patients were assessed with pain,shoulder mobility,complications,strength and the Constant score,comparing with the contralateral side.Results Among all the 106 cases,95 cases used 5 hole plate,11 cases used other types of plate.After 12 months of follow-up,matoration of injured shoulder enhanced obviously(P<0.05).Shoulder protraction can reach(132 ° ± 35 °);outward(122 ° ± 39 °);out-rotation(45 ° ± 23 °);intorsion(77 ° ± 21 °).One year postoperation,the Constant score was(70.6±13.7),(85.1%±14.0%) of the contralateral side(83.0±8.0).In addition,the score enhanced as the follow-up time went on(P<0.05).Among the 93 patients who finished the follow-up,25(27%) patients appeared 34 complications.18 complications(53%) happened in 3 months post-operation.22 complications(65%) were related to incorrect surgical technique.Conclusion Surgical treatment of displaced proximal humeral fractures using locking proximal humeral plate can lead to a good functional outcome and most of the complications were related to incorrect surgical technique.

    Objective To evaluate the clinical results of open reduction and internal fixation combining bone graft in the treatment of humerus surgical neck fracture(NeerⅡ,Ⅲ).Methods Twenty-one cases of humerus surgical neck fractures(Neer Ⅱ: 5 cases;Neer Ⅲ: 16 cases) were treated by locking proximal humerus plate combining bone grafting.Results All patients were followed for more than 6 months on average,all of the fractures were healed and got satisfactory results of shoulder functional recovery.The results were excellent and good in 18 cases,the good rate was 85.7%.Conclusion Locking proximal humerus plate fracture fragments combining bone graft is conductive to the fracture healing and shoulder functional recovery.It is an effective method in the treatment of humerus surgical neck fracture(Near Ⅱ,Ⅲ).

    Objective To explore the effect of two injuries of superior shoulder suspensory complex(SSSC) by surgical treatment.Methods A retrospective analysis was done on 18 cases with two injuries of SSSC from August 2000 to February 2010,and the patients had different degrees of merger damage.All of the 18 cases were treated surgically,a simple immobilization was done for the obviously displaced clavicular or dislocated acromioclavicular joint in 11 cases,and combined fixation was conducted for the rest.Results According to the Herscovici system,the result was classified as excellent in 13 cases,good in 2 cases,fair in 2 cases and poor in 1 case.Conclusion The stability of the SSSC was recovered after surgery,but personalized operative treatment program should be adopted.

    The Application of Transfixing Suture in Sealing up the Dead Space Resulting from Resection of Body Surface Tumors

    Objective To explore the effect of transfixing suture in sealing up the dead space resulting from resection of body surface tumors.Methods Among 208 cases of body surface tumor,relatively huge lacuna was formed in176 cases after tumor resection.All cases were divided into two groups by manner of operation: regular layered suture technique combined with drainage(group A,n=97) and transfixing suture technology combined with pressure dressing(group B,n=79) respectively.The incidence of complications of two groups was observed and compared after operation.Results The incidence rate of complication is 7.21% in group A and 1.26% in group B.There was significant difference between group A and B(P<0.01).Conclusion Transfixing suture has much lower incidence rate of complications and shows advantages in sealing up dead space and hemostasis.It is recommendable in clinical application.

    Insulin-like growth factors(IGFs) are important growth factors regulating body growth and development especially on endochondral ossification.IGF-I involves in chondrogenesis of mesenchymal stem cells and maintenance of mature cartilage.Insulin-like growth factor-binding proteins(IGFBPs) represent a evolutionarily conservative protein family including 6 independent family members:IGFBP1,2,3,4,5,6.They regulate functions of IGFs,exert functions of storage and transport of IGFs and have some IGF independent actions.IGFBPs play an important role in regulating cartilage development too.The regulatory role of IGFs and IGFBPs on chondrocytes growth and development were reviewed.The references for cartilage tissue engineering were provided.

    Epigenetic reprogramming of nuclei using cell extract becomes popular in recent years.This method indicates that recipient cells receiving transduction of cell extract from donor cells can generate changes of morphology,gene expression and function directing donor cells.It is reported to be used between somatic cells and somatic cells or stem cells as an effective method with no ethical controversy.The current situation,relevant mechanisms and defects concerning reprogramming of cell extract were reviewed.
